pDefend2 is a plasmid that that encodes for a mutant of yeast-phe tRNA. It was constructed by restricting pDEFEND2 with BSA I and BstBI, followed by ligation of the appropriate sequence to the unique sticky ends. In this mutant the modifications of yeast-phe tRNA have been removed.

pDEFEND2 is cut with BsaI and BstBI in order to "open up" the plasmid for insertion of the desired template. BsaI removes four essential bases (ATAG) from the T7 promoter region and therefore must be added to the plus strand of the template.
