Problem Set on DNA Forensics
1) Consider the assigned papers on the stabilities of various mismatches
and on the methods employed in DNA forensics.
(a) Invent a circumstance (with specific sequences of probes/primers and targets)
in which mis-hybridization could cause ambiguity or
error in data interpretation or conclusions.
(b) Propose a method with which one could estimate probabilities of
such errors and ambiguities.
2) From the Wartell paper on TGGE:
(a) Each lane in Figure 2 contains several bands.
From a discussion of the PCR reactions, the annealing reactions, etc., explain the
origin of each of these bands.
(b) The authors note that the mismatches are in the lowest melting region of the DNA.
Does that matter in this experimental method (yes)? Why?
